Amine-activated SepFast MAG is used to make magnetic chromatography adsorbents. It forms chemically stable amide linkages with ligands containing carboxyl groups. It has a long spacer (9 atoms). After binding of target molecules, the magnetic resin can be readily isolated from the biological feedstocks with the aid of a magnet.
Amine-activated SepFast MAG 4HF is made of highly cross-linked 4% agarose containing magnetic material. It makes harsher operational conditions possible, so that low pH and certain organic solvents of high concentrations can be applied.
The base matrix is activated by attaching a long hydrophilic chain with an amine group in the end. They react with either carboxyl containing ligands in the presence of a carbodiimide agent, or aldehyde containing ligands in the presence of a reducing agent. Amine-activated SepFast is supplied as an aqueous suspension in 20% ethanol.
