| For control of the optical purity of a substance, the two columns NUCLEOSIL® CHIRAL-2 and NUCLEOSIL® CHIRAL-3 allow to select conditions such that the minor enantiomer, which is present as an impurity, is eluted before the main peak. NUCLEOSIL Chiral-1配位交換手性柱
配位交換色譜LEC(ligand exchange chromatography)的分離原理是，被測手性樣品與色譜柱的官能團中的金屬陽離子（CU2+）產生三重混合配位交換，而L-羥基-脯氨酸產生的不對稱中心導致了對樣品的手性色譜拆分。NUCLEOSIL CHIRAL-1 配位交換手性柱色譜條件的選擇我們建議使用CUSO4的水溶液作為NUCLEOSIL CHIRAL-1的流動相，并可以按一下原則通過改變流動相條件改善分離效果。
Chiral-2/3刷型交換手性柱
NUCLEOSIL CHIRAL-2/3手性柱是正相原理分離手性化合物的。它是在Nucleosil硅球上交聯鍵合N-(3,5-二硝基苯酰)-“D,L”-苯基甘氨酸作為手性官能團，分離的原理是刷型交換（brush type）。這種分離原理主要用來控制農藥或藥物的生產工藝中間體的光學純度。
由于NUCLEOSIL CHIRAL-2使用的官能團是D-苯基甘氨酸，而NUCLEOSIL CHIRAL-3使用的官能團是L-苯基甘氨酸，使兩種手性柱在分離同一手性化合物時，出峰順序正好相反。這在質檢控制產品光學純度時是十分重要的，我們可以選擇NUCLEOSIL CHIRAL-2或CHIRAL-3，讓雜質在主要成份前出峰，避免由于脫尾造成定量的不準確。色譜柱使用條件：
NUCLEOSIL CHIRAL-2/3手性柱使用的流動相一般是非極性有機溶劑（正乙烷，異辛烷），添加極性有機溶劑（四氫呋喃，醇類，氯代烴）和極少量強酸（三fuyisuan）。強酸通常能夠明顯的改善手性樣品的分離。而被測的堿性樣品，需要通過苯甲酰氯或3,5-二硝基苯甲酰氯衍生化后進行檢測。
產品應用:
NUCLEOSIL Chiral-1典型應用
如果樣品中有兩個合適空間位置的極性基團，能夠與銅離子產生整合作用，CHIRAL-1就應該能夠成功地進行手性拆分。例如α-氨基酸（氨基和羧基），α-羥基羧酸（乳酸等），N-烴基α-氨基酸，以及小分子量的手性化合物。D,L-phenylalanine, D,L-pyenylglycine, D,L-proline, D,L-threonine, D,L-valine.
CU2+濃度
增加CU2+濃度可以縮短樣品的保留時間。典型的CU2+濃度應該在0.2m M-3m M之間；如下圖NUCLEOSIL CHIRAL-1在分離（D,L）-methionine蛋氨酸時，CU2+濃度的提高，分離度相應下降。 |
有機改性劑
加入有機溶劑（如甲醇：乙腈）可以縮短樣品的保留時間，提高出峰對稱性。 |
溫度
提高溫度可以相應提高柱效，縮短樣品的保留時間，建議一般使用溫度60℃，zui高的柱壓不超過140bar。如下圖NUCLEOSIL CHIRAL-1在分離D,L-alanine丙氨酸時，較低的溫度，可以提高樣品的分離度。
